Formulation and Cycle Design

  • Are the critical formulation properties (collapse temperature, eutectic point, glass transition Tg') characterized? *
  • Is the cycle designed with defined freezing, primary drying, and secondary drying stages? *
  • Are the critical process parameters (shelf temperature, chamber pressure, time, condenser temperature) defined with ranges? *
  • Are target residual moisture and cake appearance acceptance criteria defined? *

Equipment Qualification and Controls

  • Is the lyophilizer qualified, including shelf temperature mapping and empty/loaded chamber studies? *
  • Is chamber pressure control and pressure measurement (capacitance and thermal) calibrated and cross-checked? *
  • Is condenser capacity confirmed sufficient for the maximum sublimation load? *
  • Is steam sterilization (SIP) of the chamber validated between batches? *

Sterility Assurance and Loading

  • Is the loading and unloading of partially stoppered vials performed under Grade A protection? *
  • Is stoppering under vacuum or inert gas controlled and verified? *
  • Are media fills designed to simulate the lyophilization hold and loading process? *

Monitoring and Batch Review

  • Are product temperature probes or process analytical tools used to confirm the drying endpoint?
  • Is residual moisture tested on finished vials and within specification? *
  • Are cycle excursions (pressure spikes, temperature deviations) investigated for product impact? *
  • Is cake appearance and reconstitution time evaluated at batch release? *
